Creative Biolabs has launched an integrated OMV & exosome-boosted CAR-T development platform, a next-generation vesicle-engineering approach designed to investigate strategies for overcoming barriers to CAR-T activity in solid-tumor models. The announcement matters because CAR-T therapy, despite transforming outcomes in blood cancers, has yet to deliver comparable success in solid tumors, where poor T-cell infiltration, an immunosuppressive tumor microenvironment (TME), and safety concerns such as cytokine release syndrome have stalled progress.
The new platform combines two complementary nanoscale vesicle technologies—bacterial outer membrane vesicles (OMVs) and mammalian-derived exosomes—into a unified, customizable pipeline. Each vesicle type targets a distinct bottleneck. The OMV engineering arm functionally remodels the tumor microenvironment to promote CAR-T infiltration, and includes the BROAD-CAR system, in which OMVs transiently program tumor cells in vivo to display synthetic antigens recognized by a universal CAR-T construct. This offers a preclinical strategy for evaluating antigen redirection across heterogeneous tumor models. OMV surface engineering with targeting ligands further guides CAR-T homing precision, while detoxified LPS strains require characterization of immunostimulatory activity and endotoxin-related safety—a critical differentiator from synthetic liposome carriers that lack intrinsic immune-activating capacity.
The exosome engineering arm provides a biocompatible support system to sustain CAR-T activity within the suppressive TME. CAR-exosomes—exosomes directly displaying functional CARs on their surface—deliver antigen-specific tumor killing without live cell administration, with a safety profile that requires preclinical comparison with live-cell CAR-T approaches. Selected exosome systems are being investigated for transport across biological barriers, including the blood-brain barrier, and can be engineered to carry cytokines, checkpoint inhibitors, and survival signals intended to modulate CAR-T exhaustion and persistence.
A third service line equips both vesicle types with customized therapeutic payloads. Small molecules, nucleic acids (mRNA, siRNA, miRNA), and proteins can be co-encapsulated within a single vesicle for multimodal synergy. Metabolic TME remodeling formulations alleviate iron-mediated oxidative stress, while vesicle-mediated checkpoint blockade enables localized, sustained PD-1/PD-L1 inhibition directly at tumor sites—maximizing activation while minimizing systemic toxicity.
The platform's significance lies in enabling researchers to investigate multiple barriers in parallel rather than sequentially. According to a senior scientist at Creative Biolabs, integrating OMV and exosome engineering into a single platform gives researchers a dual-mechanism toolkit that doesn't just chip away at these barriers but enables parallel investigation, from stroma penetration to metabolic persistence. For research teams selecting a vesicle-engineering partner, the company advises assessing dual-modality integration, LPS detoxification data, and cargo co-loading capacity. The platform also offers antigen-independent targeting, cell-free CAR-exosome options for repeat-dosing studies, and project-specific ex vivo workflows.
